Room temperature ionic liquids have recently been very popular as potential green solvents due to their unique physical and chemical properties of negligible vapor pressure, non-flammable and thermal stability. Even though some groups have explored to replace the conventional organic solvents with ionic liquids, few studies have been conducted on biotechnology until now. In this presentation, we will talk about the application of ionic liquids in biotechnology. First, the toxicities of ionic liquids on E. coli and yeast will be presented. It was observed that most of ionic liquids tested are less toxic than the conventional solvents such as acetone, acetonitrile, and ethanol. It was also shown that ionic liquids with longer alkyl chain in cations show higher toxicity. Next, the potential use of ionic liquids will be demonstrated as reaction media for enzymatic conversion in single- or two-phase system. As an example, in lipase-catalyzed transesterification of vinyl acetate with n-butanol, conversion yields in several ionic liquids were higher than those in conventional organic solvents. For the production of sucrose laurate through the lipase-catalyzed transesterification of sucrose with vinyl laurate, the highest conversion yield was obtained in [Edmim] [Tf2N] among ionic liquids tested. The application of ionic liquids as organic modifier in RP-HPLC was exemplified in the separation of phenylalanine and tryptophane.
