A Digital Microfluidic Platform for Synthesizing ImmunoPET Tracers

Biomolecules and their engineered derivatives provide an enormous reservoir for developing highly specific Positron Emission Tomography (PET) imaging probes to visualize biological processes and detect biomarkers of interests in vivo. However, efficient radiolabeling of a delicate biomolecule still remains challenging. Using droplets generated in microfluidics, which resemble isolated, miniature batch microreactors composed of different reagents and compositions, enables large-scale screening and exerts an unprecedented degree of control over individual reaction conditions. We have developed a novel microfluidic chip capable of generating oil-free droplets and manipulating minute volume of liquids. The feasibility of on-chip radiolabeling of biomolecules was demonstrated using various biomolecules and a common 18F-tag, N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB). Consumed only small amount of precious biomolecules, the optimal reaction parameters can be obtained by screening many individual droplets (analytical-scale “batches”) with different compositions. Batches of products can be generated from one single droplet to mouse-dose scale to match the demand of radioactivity for radiotracer evaluation. Data from in vitro studies and in vivo micro-PET scans using 18F-labeled engineered antibody fragments (diabodies) will be presented.
