Appropriate supply of nutrients, oxygen, growth factors and removal of wastes etc. are an important basic issue for design and organization of cultured 3D tissue for both regenerative medicine and cell-based assays. This is not only for allow the cells to function at in vivo-comparable levels by also for reduction of the cost to obtain large- number of cells for these purposes. Chemical engineering give us the fundamentals to analyses and optimization for that issue. I would like to introduce our latest activities concerning 1) scalable iPS cell propagation and differentiation based on rotational culture of suspended aggregates and on dialysis operations for optimized nutrient supply and waste removal while keeping both exogenous and endogenous growth factors, 2) design and 3D fabrication of scalable scaffolds for large internal organs for future implantation with immediate blood vessel connections, and 3) hierarchical 3D cocultures of liver cells using oxygen-permeable plates for physiological cultured liver tissue models.
