Exploring β-Sheet Structure and Interactions with Chemical Model Systems Abstract: This talk will describe the use of chemical models of β-sheets to study the structure and interactions of β-sheets. In these artificial β-sheets, molecular template and turn units are combined with peptides to mimic the structures of parallel and antiparallel β-sheets [1,2]. Key templates and turn units include the unnatural amino acid Hao [3], which mimics a tripeptide β-strand, and δ-linked ornithine, which mimics a β-turn [4,5]. Artificial β-sheets with exposed hydrogen-bonding edges readily dimerize through edge-to-edge hydrogen bonding in chloroform solutions [3,4,6]. NMR studies of these artificial β-sheet dimers demonstrate the importance of hydrogen-bonding complementarity, size-complementarity, and chiral complementarity in molecular recognition between β-sheets [7-9]. Water soluble artificial β- sheets that fold and self-assemble have also been developed [10,11]. Ongoing studies have lead to water-soluble artificial β-sheets that bind proteins through β-sheet interactions and control peptide aggregation associated with Alzheimer's disease. References 1.
