Recently, screening of drug, cosmetics and toxicity using animal are prevented due to ethical regulation. In addition, human based screening is becoming more difficult than before. Therefore, construction of organs under in vitro environment has been greatly focused, and the study to establish model, which can replace the function of human’s or animal’s organs, as similar as possible are being performed extensively. Due to the recent progress in microtechnology, the microenvironments surrounding cells can be precisely regulated, and such organ models called ‘organ on a chip’, which mimic the function of real organ, can be created. In this presentation, several platforms which replace organs, such as liver and brain, will be introduced. Cells were obtained from fetus of rat, and by seeding these cells to the microplatform; 2D and 3D organ models were developed. Using the proposed organ on a chip, we have tested liver toxicity of anticancer drug and wound healing of liver and studied the effect of hepatic stellate cells to hepatocyte tissue. In addition, we have made Alzheimer’s disease model and tested amyloid-β effect to networked neurons. The ‘organ on a chip’ could be used for the screening diverse products including drug, food, and cosmetics, and toxicity screening is another big application. In medicine, this can be used as disease model to reveal the mechanism of critical disease and will broaden its application extensively in near future.
