In vivo Optical imaging technologies rely on light producing optical reporters such as luciferase and fluorescent proteins, fluorescent dyes and conjugates. Genes encoding luciferase and fluorescent proteins can be engineered into cells and animals to enable them to produce light that can then be visualized through the tissues of a live animal using specialized imaging equipment and software. In this lecture, the physics as well as the instrumentation stories behind these technologies will be briefly discussed. Advanced analysis methods that provide improved sensitivity, including spectral unmixing, transmission normalized fluorescence, 3D bioluminescence tomography and fluorescence tomography, will be presented. |
