Abstract
The human nervous system contains two major divisions. The central nervous system includes the brain, the spinal cord and the eye. The peripheral nervous system includes the motor, sensory and autonomic nerves that connect with all other parts of the body. The injured central nervous system cannot repair itself, and the functions of the damaged parts of the central nervous system do not recover their function. In contrast, the peripheral nervous system has a greater capacity for repair and restoration of function. We are beginning to understand some of the biological process that allow regenerative repair in the peripheral nervous system and that prevent regenerative repair in the central nervous system. For example, injured nerve cells in the peripheral nervous system change their patterns of protein synthesis after injury to support growth, but injured nerve cells in the central nervous system do not. Also, the injured peripheral nervous system provides a supportive environment for nerve cell growth, but the injured central nervous system contains growth inhibitory factors. Tissue repair scaffolds offer an opportunity to provide(1) a structural substrate for regenerative growth, (2) delivery of 'growth factors' that can stimulate the synthesis of proteins necessary for growth, and (3) incorporation of precisely placed support cells that can create an environment favorable for growth and repair of all parts of the nervous system.
