High-throughput single-cell analysis is a challenging task. I will be shown that Label-free tomographic phase microscopy is an excellent candidate to perform this task. We demonstrate that by exploiting the random rolling of cells while they are flowing along a microfluidic channel, it is possible to obtain in-line phase-contrast tomography, if smart strategies for wavefront analysis are adopted. Without a priori knowledge of the three-dimensional position and orientation of rotating cells 3D tomography can be completely retrieved through digital holography and intelligent wavefront numerical analysis. The intriguing and challenging result is that by our approach a continuous-flow cytotomo-graphy is possible thus opening the route for practical operation in real-world, single-cell analysis.

Dr. Pietro Ferraro is director of research at CNR Institute of Applied Sciences and Intelligent Systems, Napoli Italy. He has published 12 book chapters, 300 papers in journals, 250 papers at conferences. He has edited two books with Springer and holds 14 patents. His current scientific interests are holography, interferometry, microscopy, fabrication of nanostructures, ferroelectric crystals, optical fiber sensors, fiber bragg gratings, nano-microfluidics, optofluidics, etc. He is SPIE Fellow and OSA Fellow. According to Google Scholar he has more 11.000 citations and H-index of 56.
