A membrane/core type nanoparticle delivery system will be discussed in the talk. The core consists of condensed DNA (LPD) or calcium phosphate amorphous precipitate (LCP). Either cores encapsulate siRNA or small molecular drugs with high efficiency. Both can resist uptake by reticuloendothelial system due to the presence of a PEG brush on the surface membrane. LCP de-assembles in the acidic endosome and releases its cargo to the cytoplasm. Additional nuclear targeting will require the presence of a peptide. Their applications for the therapy of cancer and liver diseases will be shown. |
